Quarterback Brock Purdy and the team's top two active wide receivers, Ricky Pearsall and Jauan Jennings, have all been ruled out for the Week 5 clash.
The news leaves the 49ers dramatically depleted for a crucial game against their division rivals.
The unexpected absences are a major blow to the 49ers' offence.
With the team already without star defensive end Nick Bosa and tight end George Kittle, the short week proved too difficult for the injured players to overcome.

The three crucial players ruled out for the Rams clash
The three major offensive contributors have all been officially ruled out of the game.
Brock Purdy (Quarterback): The 25-year-old quarterback is out with a lingering toe injury that previously sidelined him for two games. Purdy returned in Week 4 but experienced soreness after the loss to the Jacksonville Jaguars, making the short turnaround impossible.
Ricky Pearsall (Wide Receiver): The wide receiver, who leads the team in receiving yards this season, is sidelined with a knee injury.
Jauan Jennings (Wide Receiver): The 28-year-old wideout is out with injuries to his ankle and ribs, marking his second absence of the regular season.

What the absences mean for Mac Jones and the offence
The loss of three major contributors puts significant pressure on backup quarterback Mac Jones, who will be making his third start of the season.

The former first-rounder is 2-0 as a starter this season, but he will be forced to operate with a severely depleted receiving corps.
Jones will have to rely on wide receivers Kendrick Bourne and Demarcus Robinson to step up in the passing game.
The team is also expected to lean heavily on running back Christian McCaffrey and the running game to control the clock and keep the ball out of the hands of the Rams' offence.
